Nice question. I guess nobody knows the answer yet. Flow cytometry might not be the best option.

The reason plastic adherence is the method of choice till date, since few papers published using antibodies have not presented any significant advantage over conventional method.

But then at what passage does plastic adherence give a pure enough population to be considered MSCs? 5? 10? 50?

For human cells, MSCs from bone marrow are effectively purified in a single step by plastic adherence. No need for multiple passages to get rid of the hematopoietic cells.

For mouse cells, isolating MSCs from bone marrow is less simple because the hematopoietic cells are stickier, so often immunodepletion is necessary. However, my understanding is that adipose-derived mouse MSCs are easier than marrow-derived cells to isolate by plastic adherence.

From one I've read, it's easy to sort based on adherence within just a few days, usually before one passage (at least with human ADSCs). The marker profile changes very quickly.
